Protective Effect of Polysaccharide from Sargassum horneri on H2O2-induced Oxidative Stress in HaCaT Cells

    Abstract:

    To investigate the protective effect of Sargassum horneri polysaccharides (SHP) on H2O2-induced oxidative stress injury of human keratinocytes (HaCaT), the total antioxidant capacity (T-AOC), DPPH free radical, hydroxyl free radical (·OH) and superoxide anion free radical (O2·) were determined, and the antioxidant capacity of SHP in vitro and established the oxidative damage model of HaCaT cells induced by H2O2was evaluated. The protective effect of SHP on oxidative damage of HaCaT cells was evaluated by measuring cell survival rate, reactive oxygen species and enzyme activity. The results showed that the clearance rate of DPPH was 68% and the scavenging capacity of ·OH was 65.48 U/mL treated with 1 mg/mL SHP. The clearance capacity of O2· was 84.86 U/mL and T-AOC was 33.55 treated with 3 mg/mL SHP. SHP could significantly improve the viability of HaCaT cells by H2O2-induced oxidative damage. After, The survival rate of HaCaT cells increased from 56.85% to 80.57%, as well as the intracellular SOD activity treated with 100 μg/mL SHP, while the contents of ROS and MDA significantly reduced (P<0.05). Therefore, SHP had a protective effect on H2O2-induced oxidative stress injury of HaCaT cells, suggesting that SHP, a natural antioxidant, could be widely applied to cosmetics and cosmeceuticals.
