Abstract:In order to understand the function of vinorine synthase gene (VS) in Salvia miltiorrhiza, the SmVS gene were cloned from S. miltiorrhiza, and the bioinformation of SmVS was predicted by Ex PASy and other online analysis software, the expression pattern of SmVS was analyzed by RT-qPCR. The results showed that the full-length of SmVS was 726 bp, encoding 241 amino acids. The molecular weight of SmVS was 26 861.72 with isoelectric point of 5.66. It was an extramembrane protein, presumably located in mitochondria. Phylogenetic tree showed that SmVS had close relationship with VS in Olea europaea var. sylvestris. The expression of SmVS in roots was higher than that in leaves, and the transcription level of SmVS reached the highest at 21:00 pm in leaves. Therefore, it was suggested that SmVS could be involve in responsing to light.

Abstract:In order to understand the function of transcription factor WOX in Dendrobium officinale, the WOX family members in D. officinale were identified by whole genome analysis techniques, the bioinformatics of WOXs and expression patterns of WOXs were analyzed. The results showed that there were nine WOX transcription factors in D. officinale genome, named DoWOX 1 to DoWOX 9, respectively. The most of DoWOXs contained 2-3 exons, while DoWOX9 had only one exon. The promoters of DoWOXs contained cis-regulatory elements related to hormone induction, stress responsive, plant growth and development. The expression of DoWOX1, DoWOX2, DoWOX3, DoWOX4 and DoWOX9 were the highest at protocorm-like body (PLB) stage by qPCR, which of DoWOX4 was the highest at small flower bud stage, and then decreased. In addition, the expressions of DoWOX3, DoWOX7 and DoWOX9 were the highest in columns, and that of DoWOX4 was the highest in lips. Therefore, DoWOXs might be involved in regulating the growth and development of D. officinale, and play an important role in maintenance of PLBs state and flower development.

Abstract:In order to screen appropriate reference gene for real-time qPCR (qRT-PCR) in Ananas comosus var. bracteatus at different development stages, the expression stability of 10 housekeeping genes, such as EF1, UBQ, ACT, GADPH, Histone, TUA, TUB, 18S, elf-5a, and α-tubulin, were detected, and the expression of PetF in seedling of A. comosus var. bracteatus was studied. The results showed that the expression stability of these genes in albino and green seedlings was different at development stages. Histone and α-tubulin were optimum reference genes at different development stages. Compared between green and albino seedlings, the ideal reference gene combination were 18S, EF1 and α-tubulin. The expression of PetF gene had the same variation trends in green and albino seedlings at three development stages by using screening reference genes. So, it was confirmed that the reference genes screened were suitable for qRT-PCR of A. comosus var. bracteatus.
