Seed Germination in vitro of Renanthera imschootiana Rolfe

    Abstract:

    Asymbictic germination of an endangered orchid, Renanthera imschootiana Rolfe was achieved through sowing the seeds after pollinating for 145 d on different mediums axenically. In 30 d after sowing, the seed germination rate reached 93.1% on the 1/4MS medium supplemented with 20% (v/v) coconut milk(CM), 1 g L-1 peptone and 10 g L-1 sucrose. After 75 d, on the VW medium with 10% CM, 36.6% of the seed formed plantlets. The content and concentration of multi-salt of basic medium has a positive (or great) effect on seed germination and plantlet growing. 1 g L-1 peptone increased seed germination rate, while 150 g L-1 apple homogenate can enhance plantlet growth. Seed germination rate was found no significant difference when seeds were sowed in light or dark, but 3-weeks culture in dark had a negtive effect on developing seedlings after seed germination.
