Abstract:In order to establish an efficient and rapid propagation system for cuttings of Euryodendron excelsum, the effects of substrate type and soaking time of plant growth regulator on rooting of cuttings were studied by using 1-year-old lateral bud and 2-year-old lateral bud of 3-year-old tissue culture seedlings and 1-year-old lateral bud of adult seedlings. The results showed that the rooting mode of cuttings was mixed rooting type, the rooting rate of tissue culture seedlings was significantly higher than that of adult plants. The rooting rate of cuttings treated with indolebutyric acid (IBA) was much higher than that treated with naphthalene acetic acid (NAA) on medium with the volume ratio of vermiculite to perlite for 2:1. The rooting rate and average root length of cutting tissue culture seedlings soaked in 1 500 mg/L IBA for 20 min were the highest, which were 96.67% and 0.97 cm, respectively. The rooting rate of adult cuttings soaked in 2 000 mg/L IBA for 10 s was the highest (34.44%), the average root number was 4.22, and the average root length was 0.53 cm. The rooting rate was the highest (46.67%) in the top 1-3 segments of the 2-year-old lateral branches. So, the tissue culture seedlings as cuttings mother plant had a high success rate, could improve the reproduction efficiency and reduce the production cost.
